With the Champions' League deadline just one day away, Glenn is hoping Paolo can put the speculation behind him and help the Hammers to three points at Southampton's new stadium.
Manchester United boss Sir Alex Ferguson has admitted business is only going to be done if he can sell Dwight Yorke to Middlesbrough - which, at this stage, is looking unlikely.
And Glenn hopes he is able to focus on the game in hand, saying: "He is a West Ham player, he is very much in our plans and I'm expecting him to have a very good game, and hopefully play a big part helping us to get the result we want.
"We are all looking forward to the game. We had a good victory against Southampton earlier in the season in what was a hard game.
"We scored both goals in the second half, it was a good victory for us and obviously we are trying to do the same tonight."
That victory came at a time when the media spotlight was turned on Glenn and then-Saints boss Stuart Gray - and shortly after the Hammers' win Gray was sacked.
"I think everyone felt Stuart Gray was very unfortunate to be given such a short period of time to sort the job out. He didn't even get the chance to get his feet under the table before he was moved on, but it is a reflection of how precarious professional football is," says Glenn.
"We know that before we come into it, but Gordon Strachan is there now, and he is doing a terrific job.
"He has really got the players fired up, so we are going to have to meet them head on.
"Southampton have had some terrific performances of late and some good results; in a short period of time they took four points off Liverpool, they took Manchester United all the way and were quite unfortunate to lose when they made several good chances, and we saw them before Christmas at Leeds when they took them all the way and lost 1-0.
"They will be very fired up and we need to match them all the way to get the result we are looking for.
"We are now well into the second half of the season and it is important we start picking up points all the way in now."
Of the team news, Glenn adds: "John Moncur is available again and I'd be very confident of Trevor Sinclair being fit and available for Saturday's home game against Blackburn."
He hopes the players can continue the optimism gained by the draw at Stamford Bridge at the weekend, and he says: "It was a very happy dressing room after the game, obviously, after scoring a late equaliser in such an important match. We are looking forward to the replay but we have got two very, very, important league games in the next few days.
"As Alex Ferguson said last night, from now on at this time of the year there is no messing about, and what he said applies to everyone, wherever you are in the league - every team needs to pick up points as quickly as they can.
"Whoever goes out for us has to be equally as fired up as the Southampton players. We have to play our football and hopefully we will make some chances and take them when we do.
"There are no easy games in the Premiership. Every game takes an awful lot of winning and this will be no different."
Vladimir Labant could start after making his debut on Saturday as a substitute, and Glenn says: "Vlad didn't do anything wrong when he came on and he is getting closer to full fitness. He is not quite there yet but he will certainly figure in the game tonight."
As for suspensions, he adds: "Tomas Repka is going to miss one game, the Bolton game, and we are going to miss Paolo for three games. That is worrying with this small squad that we operate with, but today our focus is only on getting a result at Southampton."
He admits it is all quiet on the transfer front, despite reports linking the Hammers with Sheffield United's Michael Brown and Boca Juniors' Rolando Schiavi.
"Anything we have been linked with, there is no truth in and, as I always say, we are working hard to bring in a couple of loan signings in between now and the end of the season with the emphasis on quality.
"That is what slows everything up, finding players of sufficient quality for the 14 games after tonight."
And he denies suggestions that wrapping up the signature of 17-year-old Liam Rosenior is imminent.
"They have been slow to really get down to any serious talking so there is nothing happening there at all, to be honest," he says.
Michael Ferrante, meanwhile, has another trial game for Bristol Rovers reserves against Northampton before they decide whether to take him on a month's loan.
